De ALLG
MM6-studie (2002-2005) heeft laten zien dat consolidatietherapie met
thalidomide plus prednisolon resulteerde in verbetering van de progressievrije
en overall survival in patiënten met multipel myeloom na autologe
stamceltransplantatie. Prof. Andrew Spencer (Monash University, Melbourne) en
collega’s publiceren online in The Lancet Haematology de vijf-jaars follow-up uitkomsten van ALLG
MM6.1 In de studie waren 269 patiënten
met nieuw gediagnostiseerd MM na transplantatie gerandomiseerd naar doorlopende
prednisolon-onderhoudstherapie of prednisolon-onderhoud in combinatie met één
jaar thalidomide-consolidatie.
Na mediaan
5,4 jaar follow-up bedroeg de vijf-jaars PFS 27% (95%-bti 23-32) in de
thalidomidegroep versus 15% (11-18) in de controlegroep (HR 0,16; p=0,0054). De
vijf-jaars OS was 66% (95%-bti 61-70) in de thalidomidegroep versus 47% (42-51)
in de controlegroep (HR 0,12; p=0,0072). Er waren geen verschillen tussen beide
groepen in overleving na progressie en incidentie van secundaire maligniteiten.
Thalidomide-consolidatie kostte AUS$ 26.996,- per gewonnen levensjaar (€
18.357,-).
