De Simon two-stage study includeerde 37 patiënten met niet-eerder behandeld, PD-L1 laag of onbekend, stadium IV SQCLC die gemcitabine 1000 mg/m2 plus ABP 100 mg/m2 eens per week kregen; in stadium één 3 week on, 1 week off; en in stadium twee 2 week on , 1 week off. Het primaire eindpunt was percentage patiënten met objectieve respons. Onder de 32 voor respons evaluabele patiënten werd respons gezien in 18 patiënten (ORR 59%; 95%-bti 42-74), onder wie één patiënt met complete respons. De mediane progressievrije overleving was 7,5 maanden (95%-bti 6,7-10,5). Er waren geen onverwachte toxiciteiten.
De onderzoekers concluderen dat de combinatie van gemcitabine plus ABP een veilige, goed-verdragen, en werkzame therapie was voor nieuw-gediagnostiseerd stadium IV SQCLC.

Summary: A phase 2 study at Memorial Sloan Kettering Cancer Center (New York) found that the combination of gemcitabine and albumin-bound paclitaxel was a safe, tolerable, and effective therapy for newly diagnosed stage IV squamous cell lung cancer. The ORR was 59%, including complete response in 1 of 32 evaluable patients.
