In de PROCLAIM-database (een register van meer dan veertig tertiaire centra) identificeerden de onderzoekers 40 mM- en 17 mRCC-patiënten die HD-IL2 kregen na PD1- of PDL1-remmers, en 1122 mM- en mRCC-patiënten die HD-IL2 kregen zonder eerdere immuuncheckpoint-blokkade (ICB). De beste overall response rate op HD-IL2 in de patiënten met eerdere ICB was 22,5% voor mM (4 complete responsen, 5 partiële responsen) en 24% voor mRCC (2 CR, 2 PR). De toxiciteit van HD-IL2 was manageable, en niet verschillend van die in patiënten zonder eerdere ICB. Er waren geen significante verschillen in overall survival of progressievrije overleving tussen patiënten met of zonder eerdere ICB.
De onderzoekers concluderen dat HD-IL2 resulteert in duurzame antitumor-activiteit in patiënten met mM en mRCC, ook na eerdere ICB.

Summary: Metastatic melanoma and renal cell carcinoma are often treated with anti-PD-1 based therapy; however, not all patients respond and further therapies are needed. An analysis of the PROCLAIM database found that treatment with high-dose interleukin-2 after anti-PD-1 based therapy resulted in response in almost one in four patients. The overall survival and progression-free survival were not different between groups of patients with and without previous immune checkpoint blockade.
