SWOG 80405 randomiseerde patiënten met gevorderd CRC naar eerstelijns bevacizumab versus cetuximab, beide toegevoegd aan chemotherapie. Voor de nu gepubliceerde analyse bepaalden de onderzoekers het CMS van 581 deelnemers aan de studie, en onderzochten de associatie van het subtype met overall survival en progressievrije overleving. De analyse wees uit dat de CMSs sterk prognostisch waren voor OS (p<0,001) en PFS (p<0,001). De CMSs waren ook predictief voor de impact van beide behandelingen op OS (p voor interactie <0,001) en PFS (p voor interactie 0,0032). In het CMS1-cohort hadden de patiënten in de bevacizumab-arm significant betere OS dan patiënten in de cetuximab-arm (p<0,001). In het CMS2-cohort hadden de patiënten in de cetuximab-arm signficant betere OS dan patiënten in de bevacizumab-arm (p=0,0046).
De onderzoekers concluderen dat de analyse klinische bruikbaarheid van CMSs heeft laten zien.

Summary: An analyses of the results of the phase 3 study SWOG 80405 (bevacizumab versus cetuximab, added to first lin chemotherapy for advanced colorectal cancer) found that consensus molecular subtypes of CRC are highly prognostic for overall survival (p<0.001) and progression-free survival (p<0.001), and predictive for both OS (p for interaction <0.001) and PFS (p for interaction = 0.0032). In the CMS1 cohort bevacizumab treatment was associated with better outcome, while in the CMS2 cohort cetuximab treatment was associated with better outcome.
