Patiënten in expansiecohort B (n=144) kregen oraal osimertinib 80 mg en savolitinib 600 mg eens per dag. Patiënten in expansiecohort D (n=42) kregen osimertinib 80 mg plus savolitinib 300 mg eens per dag. Primaire eindpunten waren veiligheid en tolerabiliteit. Adverse events van graad 3 of hoger werden gezien in 57% van de patiënten in cohort B en 38% van de patiënten in cohort D. Ernstige AEs werden gerapporteerd voor 45% in cohort B en 26% in cohort D. Twee patiënten in cohort B overleden, mogelijk gerelateerd aan de behandeling. Secundair eindpunt was het percentage patiënten met objectieve respons. Deze werd gezien in 48% (95%-bti 39-56) van de patiënten in cohort B en 64% (95%-bti 46-79) van de patiënten in cohort D.
De onderzoekers concluderen dat de combinatie van osimertinib en savolitinib een acceptabel risk-benefit profile had en bemoedigende antitumor-activiteit voor gevorderd NSCLC met MET-amplificatie en EGFR-mutatie, na progressie op een eerdere TKI.

Summary: Two expansion cohorts of the multinational phase 1b study TATTON found acceptable risk-benefit profile and encouraging antitumor activity of the combination of the EGFR TKI osimertinib and the MET TKI savolitinib for MET-amplified, EGFR mutation-positive, advanced NSCLC, with progression on a previous EGFR TKI.
