RESPONSE-2 werd uitgevoerd in 48 centra in tien landen in Azië en Europa, en in Australië en Canada. De patiënten kregen ruxolitinib of BAT gedurende ten hoogste 80 weken. De BATs waren hydroxyurea, interferon of gepegyleerd interferon, pipobroman, lenalidomide, of geen behandeling. Tussen week 28 en week 80 gingen 58 (77%) van de BAT-patiënten over op ruxolitinib. Er waren 97 patiënten die ruxolitinib kregen tot en met week 260 (59 patiënten in de ruxolitinibgroep en 38 patiënten na crossover). Gedurende vijf jaar follow-up bleef het mediane hematocrietniveau onder de patiënten in de ruxolitinibgroep lager dan 45%. Zestig flebotomieprocedures waren vereist onder 74 ruxolitinib-patiënten in 260 weken, versus 106 onder 75 BAT-patiënten. Het vijf-jaars overall survival percentage was 96% (95%-bti 87-99) in de ruxolitinibgroep en 91% (80-96) in de BAT-groep. Ruxolitinib werd goed verdragen.
De onderzoekers concluderen dat deze resultaten gebruik van ruxolitinib als tweedelijns therapie voor patiënten met inadequaat-gecontroleerde PV zonder splenomegalie steunen.
