De studie includeerde patiënten in de leeftijd van achttien tot en met zeventig jaar, een WHO performance status 2 of beter, die autologe hematopoïetische stamceltransplantatie zouden ondergaan voor NDMM. De patiënten kregen drie zes-weekse inductiecycli met hetzij isatuximab-lenalidomide-bortezomib-dexamethason (‘isatuximabgroep’) of alleen lenalidomide-bortezomib-dexamethason (‘controlegroep’). Het primaire eindpunt van de nu gepubliceerde analyse was minimale residuele ziekte (MRD)-negativiteit.
De studie includeerde 660 patiënten (mediane leeftijd 59 jaar; 38% vrouwen). De isatuximabgroep telde 331 patiënten en de controlegroep 329. MRD-negativiteit na inductietherapie werd gezien in 50% van de patiënten in de isatuximabgroep versus 36%van de patiënten in de controlegroep (OR 1,82; p=0,00017). Tenminste één graad 3 of 4 adverse event werd gerapporteerd voor 63% in de isatuximabgroep versus 61% in de controlegroep. Twaalf patiënten overleden, onder wie één in de isatuximabgroep en vier in de controlegroep aan oorzaken die werden beschouwd als samenhangend met de behandeling.
De onderzoekers concluderen dat onder patiënten met NDMM toevoegen van isatuximab aan lenalidomide-bortezomib-dexamethason inductie resulteerde in significante verhoging van het MRD-negativiteitspercentage, zonder nieuwe veiligheidssignalen.

Summary: Part 1 of the phase 3 GMMG-HD7 trial, at 67 centers in Germany, evaluated MRD negativity rate impact of addition of isatuximab to lenalidomide-bortezomib-dexamethasone induction for transplantation-eligible newly diagnosed multiple myeloma. MRD negativity was seen in 50% of patients after isatuximab-lenalidomide-bortezomib-dexamethasone induction versus 36% after lenalidomine-bortezomib-dexamethasone induction (OR 1.82; p=0.00017). There were no new safety signals.
