De analyse includeerde 6391 patiënten die hadden deelgenomen aan tien RCTs: 573 BRAFmt (9%), 2059 KRASmt (32,2%), en 3759 tweevoudig-wildtype (58,8%). BRAFmt mCRCpatiënten hadden significant slechtere overall survival dan KRASmt mCRC-patiënten (gecorrigeerd HR 1,46; 95%-bti 1,30-1,64) en patiënten met tweevoudig-wildtype (aHR 2,14; 95%-bti 1,94-2,36). In vier RCTs die toevoegen van anti-EGFR therapie aan eerstelijns chemotherapie testten was deze toevoeging onder BRAFmt-patiënten niet geassocieerd met betere PFS (aHR 0,96; 95%-bti 0,71-1,30) of OS (aHR 0,85; 95%-bti 0,66-1,14).
De onderzoekers concluderen dat toevoegen van anti-EGFR therapie aan eerstelijns chemotherapie voor BRAFmt mCRC niet resulteert in betere werkzaamheid.
