Arm 1 includeerde 21 aPDAC-patiënten die ten hoogste 12 standaard vier-weekse cycli nab-paclitaxel plus gemcitabine chemotherapie kregen, plus intratumorale injecties van 500μl LOAd703 iedere twee weken, in drie doseringsniveaus uiteenlopend van 5 x 1010 tot 5 x 1011 virusdeeltjes per injectie (in de pancreastumor of een metastase). Primaire eindpunten waren veiligheid en treatment-emergent immuunrespons. Antitumor-activiteit was een secundair eindpunt.
De mediane duur van follow-up was 6 maanden (IQR 4-10). De meest-waargenomen aan LOAd703 toegeschreven adverse events waren koorts (67% van de patiënten), vermoeidheid (38%), chills (33%), en verhoogde leverenzymactiviteit; alle graad 1 of 2 met uitzondering van een transiënte graad 3 aminotransferase verhoging bij het hoogste doseringsniveau. Er werd geen hoogst-verdragen dosering gezien, zodat 5 x1011 virusdeeltjes in combinatie met nab-paclitaxel plus gemcitabine als veilige dosering werd vastgesteld. In 94% van de patiënten resulteerden LOAd-injecties in verhoging van de CD8+ effector memory cells en adenovirus-specifieke T-cellen. Onder de 18 voor activiteit evalueerbare patiënten hadden 8 objectieve respons (44%; 95%-bti 25-66).
De onderzoekers concluderen dat onder aPDAC-patiënten combinatie van nab-paclitaxel en gemcitabine met LOAd703 feasible en veilig was. Arm 2 van LOKON001 evalueert de combinatie van LOAd703 met nab-paclitaxel, gemcitabine, en atezolizumab.
