De mediane duur van follow-up was 8,33 jaar in de combinatietherapiegroep en 6,87 jaar in de placebogroep. De overall survival was niet-significant 20% langer in de combinatietherapiegroep dan in de placebogroep (na acht jaar 71% versus 65%; HR 0,80; p=0,06). De acht-jaars melanoom-specifieke overleving was 77% versus 72% (HR 0,78; 95%-bti 0,59-1,02). Onder de 792 patiënten met melanoom met BRAF V600E-mutatie was de OS wel significant langer met de combinatie dan met placebo (HR 0,75; 95%-bti 0,58-0,96). Recidiefvrije overleving (HR 0,52; 95%-bti 0,43-0,63) en afstandsmetastasevrije overleving (0,56; 0,44-0,71) waren significant beter met de combinatie dan met placebo. Er waren geen nieuwe veiligheidssignalen.
De onderzoekers concluderen dat de combinatie vergeleken met placebo resulteerde in niet-significant overlevingsvoordeel.

Summary: Final results of the multinational phase 3 COMBI-AD trial showed that among patients with resected BRAF V600-mutated stage III melanoma, twelve months of adjuvant dabrafenib plus trametinib was associated with better relapse-free survival and distant metastasis-free survival than placebo. Risk of death was 20% lower with the combination therapy than with placebo; this benefit was not significant.
