Het cohort telde 1991 patiënten met tinnitus en 2198 patiënten met gehoorverlies die CRT hadden ondergaan voor een pediatrische maligniteit. Tinnitus (9,4% versus 5,4%; p=5,1 x 10-4) en gehoorverlies (14,0% versus 10,7%; p=0,02) werden meer gerapporteerd door mannen dan door vrouwen. Overlevers met tinnitus of gehoorverlies) hadden verhoogde waarschijnlijkheid van duizeligheid of vertigo, gebruik van antidepressiva, en slechtere zelf-gerapporteerde gezondheid vergeleken met controlepersonen. Genome-wide association study van CRT-gerelateerde tinnitus identificeerde rs203248 als risicofactor, en GWAS van CRT-gerelateerd gehoorverlies identificeerde rs332013 en rs67522722. Deze laatste associatie werd bevestigd in een onafhankelijk cohort van overlevers.
De onderzoekers concluderen dat CRT-gerelateerde ototoxiciteit geassocieerd was met geslacht, verscheiden neuro-otologische symptomen, gebruik van antidepressiva, en slechtere patiënt-gerapporteerde gezondheid. GWAS van CRT-gerelateerd gehoorverlies identificeerde rs67522722 als risicofactor.

Summary: Analysis in the cohort of the Childhood Cancer Survivor Study investigated risk factor for cranial radiation therapy-related ototoxicity among adult survivors of pediatric cancer. CRT-related ototoxicity was associated with sex, several neuro-otological symptoms, increased antidepressant use, and poorer self-reported health. GWAS of CRT-related hearing loss identified rs67522722.
