RADICALS-HD, in 38 centra in vier landen, randomiseerde 1523 patiënten naar korte-duur ADT (n=761) of lange-duur ADT (n=762). De mediane follow-up was 8,9 jaar (IQR 7,0-10,0). Het primaire eindpunt was metastasevrij overleving (MFS). Er waren 313 MFS-gebeurtenissen: 174 in de korte-duur ADT-groep en 139 in de lange-duur ADT-groep (HR 0,773; p=0,029), met tien-jaars MFS-percentages 71,9% respectievelijk 78,1%. Graad 3 of hoger toxiciteit werd gerapporteerd voor 14% van de patiënten in de korte-duur ADT-groep en 19% van de patiënten in de lange-duur ADT-groep (p=0,025), zonder treatment-related deaths.
De onderzoekers concluderen dat vergeleken met zes maanden ADT, twee jaar ADT resulteerde in significante verbetering van de MFS.

Summary: The multinational randomized RADICALS-HD trial found that among patients receiving postoperative radiotherapy after radical prostatectomy for prostate cancer, compared with addition of 6 months of androgen deprivation therapy, addition of 24 months of androgen deprivation therapy improved metastasis-free survival.
