Adebrelimab
(voorheen SHR-1316) is een op PD-L1 gericht antilichaam. Een multicenter fase
1b-studie in China heeft perioperatief adebrelimab plus nab-paclitaxel en
carboplatine chemotherapie voor stadium II of III NSCLC geëvalueerd. Prof.

de studie in het Journal of Thoracic
Oncology.1
De studie
includeerde 37 patiënten, die preoperatief drie drieweekse cycli adebrelimab
plus chemotherapie kregen, gevolgd door resectie en nog zestien cycli
adebrelimab in 34 patiënten. Het primaire eindpunt was majeure pathologische
respons (MPR), centraal geblindeerd beoordeeld. MPR werd gezien 19 van 37 patiënten
(51,4%; 95%-bti 35,9-66,6), pathologisch complete respons in 11 patiënten
(29,7%; 17,5-45,8), en RECISTv1.1 respons in 26 patiënten (70,3%; 54,2-82,5).
Het twaalf-maands gebeurtenisvrije overlevingspercentage was 77,8% (95%-bti
54,1-90,3). Graad 3 of 4 treatment-related
adverse events werden gezien in 78,4%; er waren geen graad 5 TRAEs.
De
onderzoekers concluderen dat perioperatief adebrelimab plus chemotherapie
resulteerde in een substantieel percentage van patiënten met MPR, met
manageable toxiciteit. Op basis van deze resultaten is een fase 3-studie
gestart.

Summary: A multicenter phase 1b study in China
evaluated preoperative adebrelimab (anti-PD-L1) plus nab-paclitaxel and
carboplatin, followed by resection and adjuvant adebrelimab, in stage II or III
NSCLC. Toxicities were manageable. Over 50% of patients achieved major
pathologic response, and 30% achieved pathologic complete response. Twelve-month event-free survival rate was
77.8%.
