Het cohort telde 128 patiënten (mediane leeftijd 65 jaar; IQR 55-72) met CD20+ R/R FL en een ECOG performance status 2 of beter die tenminste twee eerdere lijnen behandeling hadden gekregen.De patiënten kregen subcutaar epcoritamab in vier-weekse cycli: wekelijks in de eerste drie cycli, tweewekelijks in cycli vier tot en met negen, en vervolgens elke vier weken tot ziekteprogressie of niet-acceptabele toxiciteit. Om het risico van cytokine release syndrome te verlagen werd in de eerste cyclus step-up dosing plus profylactisch prednisolon gebruikt. Primaire eindpunten waren centraal onafhankelijk beoordeelde respons en veiligheid.
De overall response rate was 82,0% (95%-bti 74,3-88,3) met complete respons in 62,5% (53,5-70,9). De meest-gerapporteerde graad 3 of 4 treatment-emergent adverse event was neutropenie (25% van de patiënten). Graad 1 of 2 CRS werd gerapporteerd voor 65% van de patiënten en graad 3 CRS voor twee patiënten (2%). Graad 1 of 2 ICANS werd gezien in acht patiënten (6%).
De onderzoekers concluderen dat epcoritamab monotherapie klinisch relevante activiteit en manageable veiligheid had onder patiënten met R/R FL.
